| Background | This gene encodes a protein that mediates ligand-independent transcription repression of thyroid-hormone and retinoic-acid receptors by promoting chromatin condensation and preventing access of the transcription machinery. It is part of a complex which also includes histone deacetylases and transcriptional regulators similar to the yeast protein Sin3p. This gene is located between the Charcot-Marie-Tooth and Smith-Magenis syndrome critical regions on chromosome 17. Alternate splicing results in multiple transcript variants. Immunocytochemistry: Formalin-fixed HeLa cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 min and blocked with 3% BSA-PBS for 30 min at RT. Cells were stained with ARG65813 anti-NCOR1 antibody in 3% BSA-PBS and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at RT in the dark. DAPI was used to stain the cell nuclei (blue).

Immunohistochemistry: Formalin-fixed and paraffin-embedded Human breast cancer tissue section. Antigen retrieval: Sodium citrate buffer (10mM, pH 6.0), Boiling bathing for 20 min. The section was stained with ARG65813 anti-NCOR1 antibody at RT and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
