Background Voltage-gated sodium channels are transmembrane glycoprotein complexes composed of a large alpha subunit with 24 transmembrane domains and one or more regulatory beta subunits. They are responsible for the generation and propagation of action potentials in neurons and muscle. This gene encodes one member of the sodium channel alpha subunit gene family, and is highly expressed in nociceptive neurons of dorsal root ganglia and trigeminal ganglia. It mediates brain-derived neurotrophic factor-evoked membrane depolarization and is a major effector of peripheral inflammatory pain hypersensitivity. Mutations in this gene have been associated with hereditary sensory and autonomic neuropathy type VII and familial episodic pain syndrome-3. Function This protein mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a sodium-selective channel through which sodium ions may pass in accordance with their electrochemical gradient. It is a tetrodotoxin-resistant sodium channel isoform. Also involved, with the contribution of the receptor tyrosine kinase NTRK2, in rapid BDNF-evoked neuronal depolarization. [UniProt]

    Immunohistochemistry: Paraffin-embedded Rat spleen tissue. Antigen Retrieval: Heat mediation was performed in EDTA buffer (pH 8.0). The tissue section was blocked with 10% goat serum. The tissue section was then stained with ARG42591 anti-SCN11A antibody at 1 µg/ml dilution, overnight at 4°C.

    Western blot: 50 µg of samples under reducing conditions. PC-3, Rat brain, Rat kidney, C6, Mouse spleen and Mouse brain lysates stained with ARG42591 anti-SCN11A antibody at 0.5 µg/ml dilution, overnight at 4°C.

    Flow Cytometry: U-87 cells were blocked with 10% normal goat serum and then stained with ARG42591 anti-SCN11A antibody (blue) at 1 µg/10^6 cells for 30 min at 20°C, followed by incubation with DyLight®488 labelled secondary antibody. Isotype control antibody (green) was Rabbit IgG (1 µg/10^6 cells) used under the same conditions. Unlabelled sample (red) was also used as a control.
