Application Note | In Co-IP experiment, denatured IgG from IP samples may interfere target signal in immunoblotting at ~55 kDa (heavy chain) and ~25 kDa (light chain) by using traditional secondary antibodies. ARG65668 DefineBlot Mouse anti-Rabbit IgG antibody (HRP) only detecting native IgG but not denatured IgG is easy to replace traditional secondary antibodies and useful to decrease denatured IgG interference in Co-IP assay. WB: It is suggested using less than 10 μg of primary antibody for IP process, and we also recommend freshly adding 100-200 mM DTT in loading buffer for IP sample and boiling for 5-10 min to completely denature IgG. * The dilutions indicate recommended starting dilutions and the optimal dilutions or concentrations should be determined by the scientist. |
