Application Note | IHC-P: Antigen Retrieval: Boil tissue section in 10mM Citrate buffer (pH 6.0) or EDTA pH 8 for 10-20 min followed by cooling at RT for 20 min. Or perform enzymatic antigen retrieval with pepsin at 1mg/ml Tris-HCl, pH 2.0 for 10-15 min at or trypsin at 1mg/ml TrisHCl (or PBS), pH 7.4 for 10-15 min at RT. * The dilutions indicate recommended starting dilutions and the optimal dilutions or concentrations should be determined by the scientist. |

Background | Cytokeratin 19 is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21.
